1. Embryogenic calli can be initiated from mature embryos on N6E30 medium.
2. Transformation and selection are the same as the procedures for maize.
3. Regeneration medium I and II are different from those for maize.
RM-1: N6 salts, N6 vits, 2g/L casamino acids, 30g/L maltose, 20g/L sorbitol, 2mg/L BAP, 1mg/L NAA, 5mg/L ABA, 3.2g/L phytagel, 30mg/L hygromycin, pH 5.8
Duration: 2-3 weeks
RM-2: N6 salts, N6 vits, 2g/L casamino acids, 30g/L maltose, 2mg/L kinetin, 0.1mg/L NAA, 6g/L phytagel, pH 5.8
Duration: 10 days
4. Rooting medium is the same for maize.
